A transcription unit consists of:
(i) A prenatal diagnostic technique where amniotic fluid surrounding the fetus is extracted to analyze fetal cells and dissolved substances. (ii) To detect chromosomal abnormalities (e.g., Down syndrome) or metabolic disorders. (iii) To prevent its misuse for illegal sex determination, which leads to female foeticide and disrupts the natural sex ratio.
(i)(a) Spermatogenesis: Spermatogonia multiply by mitosis. Some grow into Primary Spermatocytes (2n). They undergo Meiosis I to form two Secondary Spermatocytes (n). These undergo Meiosis II to form four Spermatids (n), which differentiate into Spermatozoa (spermiogenesis). (i)(b) 1. Growth phase is much longer in oogenesis than in spermatogenesis. 2. Spermatogenesis produces four functional sperms from one primary spermatocyte, whereas oogenesis produces only one functional ovum (and polar bodies) from one primary oocyte. (ii)(a) IVF (In Vitro Fertilization): Fertilization occurs outside the body in simulated laboratory conditions, followed by embryo transfer. ZIFT (Zygote Intrafallopian Transfer): Zygote or early embryo (up to 8 blastomeres) is transferred into the Fallopian tube. GIFT (Gamete Intrafallopian Transfer): Transfer of an ovum collected from a donor into the Fallopian tube of another female. (ii)(b) GIFT is recommended for females who cannot produce viable ova but can provide a suitable environment for fertilization and embryonic development inside their fallopian tubes.
(i)(a) Hershey and Chase grew some bacteriophages on a medium with radioactive phosphorus (³²P) to label DNA, and others on radioactive sulfur (³⁵S) to label proteins. They allowed these phages to infect E. coli. The mixture was agitated in a blender to separate viral coats from bacteria, then centrifuged. (i)(b) Bacteria infected by ³²P-phages were radioactive, while those infected by ³⁵S-phages were not (radioactivity remained in the supernatant). This proved definitively that it is the DNA (not protein) that enters the host cell and acts as the genetic material. (ii)(a) In the absence of lactose, the repressor protein (synthesized by the i gene) binds to the operator region, preventing RNA polymerase from transcribing the structural genes (z, y, a). In the presence of lactose (inducer), it binds to the repressor, changing its shape so it cannot bind the operator. RNA polymerase then transcribes the operon. (ii)(b) It is termed 'negative control' because the operon is normally kept switched OFF by a repressor protein until an inducer removes the repression.
(i)(a) 'Evil Quartet' includes: 1. Habitat loss and fragmentation (most important cause). 2. Over-exploitation (e.g., Steller's sea cow, passenger pigeon). 3. (i)(b) In situ conservation protects species in their natural habitats (e.g., National Parks, Biosphere Reserves). (ii)(a) Primary treatment: Physical removal of large/small particles through sequential filtration and sedimentation to form primary sludge and effluent. Secondary treatment: Biological process where effluent is aerated. Aerobic microbes grow into flocs and consume organic matter, reducing BOD. The effluent is then passed to a settling tank where flocs sediment (activated sludge). (ii)(b) Flocs are masses of bacteria associated with fungal filaments. They are essential as they vigorously consume the major part of the organic matter in the effluent, significantly reducing the Biochemical Oxygen Demand (BOD) and purifying the water.
